The Hughes Hotel, opened in 1892, became a focal point of the town and drew travelers from the Greenville-Spartanburg road. As of the 2020 United States census, there were 4,041 people, 1,323 households, and 843 families residing in the town. In the town, the population was spread out, with 33% under the age of 18, 11.0% from 18 to 24, 30.5% from 25 to 44, 14.8% from 45 to 64, and 10.7% who were 65 years of age or older. History

In 1811, a post office was established on what is now S.C. Highway 290. In 1854, the town changed its name to Vernonville or Vernonsville in honor of local physician J.J. Vernon. Within two years of its name change, Duncan boasted a population of 200, along with several general stores, saw and flour mills and a cotton gin. The Rock Hill Negro School opened in Duncan in 1881. The Hughes Hotel, opened in 1892, became a focal point of the town and drew travelers from the Greenville-Spartanburg road. The town received its charter in 1889. It became the site of an important footnote in Spartanburg County's African-American history when the Rock Hill Nigger school opened in 1886. It was named after Leroy Duncan, who was an ambitious man who craved recognition. He promised townspeople he would provide land for city streets if they would agree to rename the town after him. The name change became official in1881. It is now a suburb of Greenville, South Carolina, and is home to the University of South Carolina at Spartanburg, which was founded in 1883. The city's name was changed to Duncan after Duncan's son-in-law, Leroy, moved to the town in 1873. It has been named after Duncan since 1881, when the town was named for the town's first postmaster, who died in 1882. It's also known as Duncan or Vernonville.

Hispanic or Latino of any race were 3.41% of the population. The town's median income was $27,974, and the median income for a family was $28,547. In the town, the population was spread out, with 33.0% under the age of 18 and 30.5% from 25 to 44, 14.8% from 45 to 64, and 10.7% who were 65 years of age or older. The median age was 28 years. For every 100 females, there are 85.6 males. The per capita income for the town is $13,194. About 19.1% of families and 20.3%. of the residents were below the poverty line, including 29.4% of those under age 18 and 13.0%. The town is located on the U.S. Census Bureau's Western Slope, which includes parts of South Dakota, Montana, and New Mexico. It is the only town in the state to have been added to the United States Census Bureau in the 1990s. It has a population of 2,870.

The offices are located on the campus of the University of Duncan, where the Duncan School of Business is based, and the Duncan International Airport, which is also based in the city. The Duncan campus is also home to the Duncan College of Business, which was founded in 1881. It was the first private college in the country. It is now the second oldest college in North America, with more than 2,000 students. The campus is home to a variety of businesses, including the Duncan Business School, which began in 1883. It also hosts the Duncan University of Applied Sciences, a business incubator, and a number of other colleges and universities. It has been the home of the Duncan Chamber of Commerce for more than 50 years. The town's history dates back to the 17th century, when it was known as "Duncan Town" and was the site of the town's first business.
